Growth and cell wall changes in azuki bean epicotyls II. Changes in wall polysaccharides during auxin- induced growth of excised segments
نویسندگان
چکیده
Changes in cell wall polysaccharides and mechanical properties of the cell wall were examined during IAA-induced elongation growth of excised azuki bean epicotyl segments under different growth conditions. Sucrose promoted IAA-induced cell elongation, but had very little effect on IAA-induced cell wall loosening. In the absence of sucrose, the amount of galactose in the cell wall decreased during the incubation period. IAA enhanced the decrease in the galactose level. In the presence of sucrose, on the other hand, IAA induced increases in the amounts of cellulose, galactose and xylose in noncellulosic polysaccharides. These IAA-induced increases were not observed in the presence of mannitol at concentrations higher than 0.1 M, although cell wall loosening was induced by IAA even in the presence of 0.2 M mannitol.

Cellular basis of growth suppression by submergence in azuki bean epicotyls.
BACKGROUND AND AIMS Complete submergence severely reduces growth rate and productivity of terrestrial plants, but much remains to be elucidated regarding the mechanisms involved. The aim of this study was to clarify the cellular basis of growth suppression by submergence in stems. METHODS The effects of submergence on the viscoelastic extensibility of the cell wall and the cellular osmotic co...
متن کاملPossible involvement of 65 kda MAP in elongation growth of azuki bean epicotyls.
Although regulation of the dynamics of plant microtubules (MTs) by microtubule-associated proteins (MAPs) has been suggested, the mechanism has not yet been elucidated. As one candidate, a MAP composed of a 65 kDa polypeptide (65 kDa MAP) has been isolated from tobacco cultured cells [Jiang and Sonobe (1993), J. Cell Sci 105: 8911. To investigate the physiological role of the 65 kDa MAP in situ...
متن کاملChanges of major wall polysaccharides and glycoproteins of tobacco cells in response to excess boron
Effects of excess concentrations of boron on major cell wall components of tobacco cells(Nicotiana tabacum L. cv. Burley 21) were studied. Pectin, xyloglucan, hydroxyproline-richglycoproteins (extensin), and arabinogalactan proteins were characterized. Results showedthat increased boron supply resulted in significant decrease in cell and cell wall dry weights. Also, high concentrations of boron ...
متن کاملGradient of Growth, Spontaneous Changes in Growth Rate and Response to Auxin of Excised Hypocotyl Segments of Phaseolus aureus.
Spontaneous growth was studied in excised mung bean (Phaseolus aureus Roxb.) hypocotyl segments. Measurements were made with a growth-recording apparatus using displacement transducers on single 5- to 6-millimeter samples excised from the growth zone immediately below the hook.Even for a given zone and under controlled experimental conditions, there are differences in the spontaneous growth of ...
متن کاملTwo azuki bean XTH genes, VaXTH1 and VaXTH2, with similar tissue-specific expression profiles, are differently regulated by auxin.
To gain insight into the functional diversity of the XTH (xyloglucan endotransglucosylase/hydrolase) gene family, we analyzed the expression profiles of two azuki bean genes, VaXTH1 and VaXTH2, which share a striking resemblance in their amino acid sequences. The two XTH genes exhibit essentially similar tissue-specific expression profiles, in that both mRNAs are found predominantly in the phlo...
